摘要
目的 测定丹参原药材和丹参饮片在与不同温度的水接触后丹酚酸B的含量以及变化,并分析含量变化的原因。方法 采用高效液相色谱法,色谱柱为ZORBAX SB-C18,流动相为5%冰乙酸-乙腈(82∶18),流速为1.0 mLmin-1,柱温为25 ℃,检测波长为285 nm。结果 丹参原药材和丹参饮片在与20~70 ℃水接触后丹酚酸B的含量均有下降,且与水接触时间越长,含量下降越严重。但水温越高,含量下降幅度越小。相同水温条件下丹参饮片中丹酚酸B的含量下降幅度小于丹参原药材。结论 丹参中的某种酶类物质在一定温度和有水的条件下会水解丹酚酸B,但高温可以破坏一部分酶的活性。应尽量避免丹参原药材以及丹参饮片与水的接触。在丹参的水提取大生产中,应该采用丹参饮片投料。
Abstract
OBJECTIVE To determine the contents of salvianolic acid B in Salvia Miltiorrhiza and its decoction pieces after contact with water at different temperatures, and analyze the causes of the content changes. METHODS High performance liquid chromatography was performed on a ZORBAX SB-C18 column, the mobile phase consisted of 5% acetic acid and acetonitrile (82∶18), the flow rate was 1.0 mLmin-1, the column temperature was 25 ℃, and the detection wavelength was 285 nm. RESUTLS The contents of salvianolic acid B in Salvia Miltiorrhiza and its decoction pieces were all decreased after contact with water at 20-70 ℃, with larger decline after longer contact time. The content of salvianolic acid B in Salviae Miltiorrhizae decoction pieces at the same temperature decreased less than that in Salvia Miltiorrhiza. CONCLUSION Some enzymes in Salvia Miltiorrhiza can hydrolyze salvianolic acid B at certain temperatures in the presence of water, but high temperature can destroy part of the activity of the enzyme. Salvia Miltiorrhiza and its decoction pieces should be kept away from water. In the water extraction of Salvia Miltiorrhiza in mass production, Salviae Miltiorrhizae decoction pieces should be used.
